摘 要:目的探讨急、慢性染铅对大鼠海马cAMP反应元件结合蛋白(CREB)的影响。方法急性染铅实验:取正常30d大鼠海马脑片培养,分别用含20μmol·L-1醋酸铅及不含醋酸铅的人工脑脊液(ACSF)孵育,作为急性染铅标本。慢性染铅实验:孕鼠在体染铅,取新生大鼠海马作为慢性染铅标本。用蛋白质免疫印迹法测定标本CREB的活性(即磷酸化CREB)和总量CREB的含量。结果急性染铅实验:海马脑片培养中,染铅组CREB的活性随时间延长而下降,染铅组和对照组总量CREB含量均无显著性变化。慢性染铅实验:总量CREB的含量无显著性变化。结论急性染铅引起CREB的活性下降,对总量CREB含量没有显著影响。慢性染铅总量CREB含量无显著性变化。Objective To study the effect of acute and chronic lead exposure on cAMP response element binding protein (CREB) of rat hippocampus. Methods For acute lead exposure in vitro,rat hippocampus was cultured with 20 μmol·L^-1 lead acetate. Distilled water,instead of lead acetate was used in the control group. For chronic lead exposure in vivo, pregnant female rats were given 0.2 % lead acetate in their drinking water before breeding and throughout gestation and lactation. After weaning at postnatal 21 d,the pups were provided 0.2 % lead acetate in drinking water. Rats hippocampus was taken as a chronic sample at the end of 20 d, 40 d, 60 d and 80 d. The content of total CREB and phosphorylated CREB were detected by Western blots. Results In acute experiment, active CREB decreased remarkably. Total CREB in acute lead exposure group and the control group remained unchanged. In chronic lead exposure, total CREB content had no obvious changes. Conclusion Acute lead exposure can decrease the level of active CREB expression. Acute and chronic lead exposure have no effect on the content of total CREB.
