多巴胺对正常大鼠和内毒素休克大鼠胃肠电的影响

摘  要:目的:探讨多巴胺对正常大鼠和内毒素休克大鼠胃肠电的影响,为临床合理应用多巴胺提供实验依据。方法:Wistar雄性大鼠40只,用药前1周在大鼠的胃、十二指肠、空肠的浆膜面埋置银丝双极电极。随机分为4组(每组10只):正常对照组经颈静脉及灌胃给予相当量的生理盐水;正常多巴胺用药组经颈静脉给予2g/L多巴胺生理盐水(4.5ml·kg-1/h,30分钟);内毒素休克不用药组经颈静脉给予大肠杆菌O111B4内毒素(15mg/kg);休克多巴胺用药组在注入内毒素后即刻给予多巴胺治疗(剂量同上)。观察上述药物对胃肠电的影响。结果:多巴胺使正常大鼠的胃、十二指肠、空肠电快波频率及振幅指数明显降低(与正常对照组比较,P<0.001);内毒素休克时胃电快波频率>5次/min,胃电快波的振幅指数、十二指肠和空肠的电快波频率及振幅指数均明显降低,P均<0.001;多巴胺使休克时的胃肠电抑制得到明显改善,胃电快波频率降低到(4.34±0.41)次/min,胃电快波的振幅指数升至29.85±5.78(内毒素休克不用药组振幅指数为21.08±3.05,P<0.01)。内毒素休克用多巴胺组及内毒素休克不用药组的十二指肠电的快波频率及振幅指?Objective:To determine whether dopamine affects the gastrointestinal (GI) motility in normal and shock conditions.Methods:Forty Wistar rats were divided into 4 groups at random.Three bipolar electrodes were implanted on serosa surface of duodenum,jejunum and stomach respectively a week prior to experiment.2 g/L dopamine salt liquid was given to the normal or endotoxic shock rats at 4 5 ml·kg 1 /h for 30 minutes by intravenous administration.Endotoxic shock was induced by Escherichia coli O 111 B 4 lipopolysaccharide challenge at a dose of 15 mg/kg.Results:It was shown that the frequency and amplitude index of GI electrical activity were sharply decreased after the administration of dopamine in normal rats( P <0 001).During endotoxic shock GI electrical activity was inhibited significantly,gastric electrical activity became tachygastria,the frequency and amplitude index of fast wave in duodenum and jejunum were small compared with that of normal rats (all P <0 001).However,infusion of dopamine resulted in disappearance of tachygastria and increased the frequency and amplitude index of spike wave (compared with nontreatment group,all P <0 01).Conclusions:The results suggest that dopamine inhibits the GI motility in normal rat and improves the GI motility in endotoxic shock rats.
