局灶节段硬化性肾炎患者血清肝细胞生长因子及肾组织中β-转化生长因子水平与肾纤维化的相关性

摘　　要：目的研究局灶节段硬化性肾炎(focal segmental glomerulosclerosis,FSGS)患者血清肝细胞生长因子(hepatocyte growth factor,HGF)、肾脏组织中β-转化生长因子(transforming growth factor-β,TGF-β)与肾纤维化程度的相关性。方法选择2004年3月-2007年11月我院收治的FSGS患者80例,均行肾穿刺病理活检确诊。免疫组化SABC法检测肾组织中TGF-β水平,用放射免疫法检测肾脏组织匀浆Ⅳ型胶原(Ⅳ-C)水平,生物素-亲和素酶联免疫吸附试验(ABC-ELISA)测定血清HGF水平,对检测结果进行相关性分析。结果FSGS患者血清HGF水平与肾组织中的Ⅳ-C水平呈负相关(P<0.05);肾组织中的TGF-β水平与Ⅳ-C水平呈正相关(P<0.05)。结论血清HGF、肾组织中的TGF-β对评估肾纤维化程度有一定的参考价值。Objective To investigate the relationship of serum hepatocyte growth factor （HGF） level, transforming growth factor - β（ TGF - β） level in renal tissue with the severity of renal fibrosis in patients with focal segmental glomerulosclerosis （FSGS）. Methods A preceding study were performed in 80 inpatients with biopsy -proven FSGS in our hospital from March 2004 to November 2007. TGF - βlevel in renal tissue was measured by histo - immunochemical SABC method, collagen -IV level in renal tissue homogenate was measured by radioimmunoassay, and serum HGF level was measured by double antibody sandwich ABC - ELISA method. And correlation analysis was made for the results. Results The serum HGF level and collagen - IV level were negatively correlated （ P 〈 0. 05 ） ; and the TGF - β level in renal tissue and collagen - IV level were positively＂ correlated （ P 〈 0. 05） Conclusion Serum HGF level and TGF - β level in renal tissue are of a certain reference significance in evaluating the severity of renal fibrosis.
