摘  要:目的比较两种脐血来源内皮祖细胞的增殖和抗凋亡能力。方法采用Annexin V/PI的凋亡检测比较两种细胞抵抗凋亡的能力;用增殖实验等方法比较两种内皮祖细胞的增殖特点;用集落形成实验比较两种细胞的集落形成能力。结果发现高增殖潜能内皮祖细胞(HPP-EPCs)显示比循环成血管细胞(CACs)具有更强的抗凋亡能力。CACs只能达到4~6个倍增,而HPP-EPCs能达到60多个倍增。CACs的单个细胞不能形成集落,HPP-EPCs的单个细胞中能在14d的培养期间形成了次级集落和三级集落。结论HPP-EPCs比CACs具有更强的抗凋亡能力、增殖能力和集落形成能力。Objective To compare the biological characteristics between the two types of endothelial progenitor cells. Methods Flow cytometry was performed to analyze the apoptosis in two types of EPCs induced by 48-hour serum-free culture. The growth curves were generated and the PDTs and CPDLs were calculated for the two types of cells and colony - forming assays were performed to compare the clonogenic potential between them. Results FACS showed that the incidence of apoptosis induced by 48 h serum-free culture in HPP - EPCs is less than CACs. The proliferation potential of HPP-EPCs was significant higher than CACs as the growth curve and MTT assay shown. The experiments of colony forming showed that no colony was formed in 960 individual cells from CACs, whereas about 45 percent of 960 individual cells formed colonies with more than 50 cells in a 2-week culture period and 5 tertiary colonies were formed in the total 96 individual cells from the secondary colonies. These data suggested that different hierarchies of EPCs from human umbilical cord blood including CACs and HPP - EPCs had been cultured and the later contained HPP - ECFC and LPP - ECFC. Conclusion HPP - EPCs possesses stronger ability of proliferation, clonogenic potential and resistance to apoptosis.
