人参皂苷对烫伤脓毒症大鼠CD45^＋和白细胞介素-2水平影响的研究

摘　　要：目的:探讨人参皂苷对烫伤脓毒症后CD45+细胞和白细胞介素-2(IL-2)的影响及其作用机制。方法:健康雄性SD大鼠66只,随机分为脓毒症组(24只)、人参皂苷组(24只)和对照组(18只);将大鼠用沸水致背部30%总体表面积Ⅲ度烫伤,烫伤后12h经腹腔注射内毒素(5mg/kg)予以"二次打击",制成烫伤脓毒症模型。用流式细胞仪检测大鼠外周血中CD45+细胞的细胞数;用放射免疫分析法检测血清IL-2的含量。结果:"二次打击"后大鼠外周血中CD45+细胞比例和IL-2水平显著下降;此过程随时间而加重,各时间点与对照组比较差异均有显著性(P(0.01);用药后24h、72h人参皂苷组CD45+细胞占淋巴细胞百分比和IL-2水平显著高于脓毒症组(P(0.01)。结论:烫伤和内毒素"二次打击"后外周血中CD45+细胞比例和IL-2水平显著下降,人参皂苷可有效恢复其功能;显著改善烫伤脓毒症时细胞免疫功能受抑状态。Objective ：To discuss the effects and mechanism of ginsenosides on CD45^＋T cells and interleukin -2 （IL-2 ）of post- scald sepsis. Methods ：Sixty-six male SD rats were randomly divided into scald sepsis group （ n = 24） , ginsenosides group （ n =24）, and control group （ n = 18）. The model of scald sepsis was made by scald injury at 30% full - thickness total body surface area and administration of endotoxin （5mg/kg） into the peritoneal cavity 12 hours after scald. Tested the cell numbers CD45 ^＋ by flow cytometry; Tested serum IL - 2 level by radioimmunoassay. Results ： CD45^＋ T cells and IL - 2 level in peripheral blood of rats significantly decreased. Compared to the control group, there was great difference at each time period （ P 〈 0.01 ） ;24 hours and 72 hours after administration of ginsenosides, the proportion of CD45 ^＋ T cells in lymphocytes, and IL - 2 level were significantly higher than those in sepsis group （ P 〈 0.01 ）. Conclusion ：The proportion of CD45 ^＋ T cells and IL- 2 level in peripheral blood of rats significantly decreased after scald and administration of endotoxin. Ginsenosides can effectively recover their function, and obviously improve the inhibit condition of the cells during scald sepsis.
