绵羊体外受精囊胚玻璃化冷冻保存的研究

摘  要:以常规的慢速冷冻法为对照,研究了玻璃化冷冻法保存绵羊体外受精囊胚的效果。对于绵羊体外受精囊胚,两种冷冻方法间的囊胚存活率没有明显差异,但玻璃化冷冻组的囊胚孵化率显著高于慢速冷冻法组(分别为76.8%和41.0%,P<0.05)。将冷冻解冻后的绵羊囊胚移植后,玻璃化冷冻组的怀孕率和产羔率明显高于慢速冷冻法的。同时比较了绵羊体外受精囊胚细胞取样后,经不同时间培养后的玻璃化冷冻保存效果。囊胚取样后培养2 h或5 h,胚胎冷冻解冻后的存活率和孵化囊胚率显著高于对照组(分别为75.0%、50.0%,78.8%、69.7%和41.2%、26.5%,P<0.05)。培养10 h后,胚胎冷冻解冻后的存活率和孵化囊胚率有所降低。结果表明,应用玻璃化冷冻法可以较好的冷冻保存绵羊体外受精囊胚及切割取样后的囊胚,绵羊体外受精囊胚切割取样后,经过2~5h时的恢复培养可以提高其抗冻能力。Vitrification and slow freezing of sheep IVF blastocysts were compared in this study. The blastocyst survival rates were similar between vitrification group and slow freezing group, but the hatched blastocyst rates were significantly higher after vitrification compared with slow freezing (76.8% and 41.0% respectively, P〈0.05). After embryo transfer, the pregnancy and lambing rates within vitrification group were higher than that of slow freezing group. Vitrification of sheep blastocysts biospied at different intervals were studied. Higher survival rate and hatching blastocyst rate were obtained after culturing 2- 5 hours than that of control(75.0%, 50.0%, 78.8%, 69.7% and 41.2%, 26.5% respectively, P〈0.05). The survival rate and hatched blastocyst rate were decreased after biospied blastocyst culturing 10 hours. In conclusion, sheep IVF blastocysts and biospied blastocysts can be vitrified successfully, and vitrification of biospied blastocysts was improved after culturing 2-5 hours.
