益气养阴活血化瘀方对实验性病毒性扩张型心肌病小鼠心肌穿孔素mRNA表达的影响

摘　　要：目的:观察益气养阴:活血化瘀方对病毒性扩张型心肌病模型小鼠心肌穿孔素(perforin,PFP)mRNA表达的影响。方法:采用Bacb/c小鼠多次腹腔注射柯萨奇B3m病毒的方式,复制重复感染的病毒性扩张型心肌病动物模型。观测心肌病理变化并以RT-PCR法检测益气养阴、活血化瘀方对病毒性扩张型心肌病小鼠心肌穿孔素mRNA表达的影响。结果:益气养阴、活血化瘀方可明显降低病毒性扩张型心肌病小鼠心肌穿孔素mRNA的表达,并能减轻其心肌病理形态学及心肌细胞超微结构的损伤。结论:益气养阴、活血化瘀方能减轻病毒性扩张型心肌病小鼠的病理损伤,抑制心肌细胞肥大,下调心肌穿孔素mRNA表达,从而对病毒性扩张型心肌病起到一定的防治作用。Objective：To observe the effect of the formula of supplementing qi and nourishing yin,promoting blood circulation to dispel blood stasis on the expression of PFP mRNA in the mice model＇with viral dilated cardiomyopathy. Methods： Replicated the animal models of the viral dilated cardiomyopthy with the method of injecting the coxB3m virus to the peritoneal cavity of the Balb/e mice. To observe the pathomorphology, PFP mRNA expression in the myocardial tissue with RT- PCR. Results ：The for- mula of supplementing qi and and nourishing yin,promoting blood circulation to dispel blood stasis could significantly reduce the PFP mRNA expression, it also could significantly alleviate the path ogenic damage of myocardial cells. Conclusion ：The formula of supplementing qi and and nourishing yin,promoting blood circulation to dispel blood stasis has significantly effect in antagonizing immune injury for Balb/c mice myocardial cells infected with coxB3m virus , the mechanism of its immunosuppression could by through lowering the PFP mRNA expression.
