氯诺昔康对手术致痛大鼠脊髓Fos蛋白表达的影响

摘  要:目的研究氯诺昔康对手术致痛大鼠脊髓Fos蛋白表达的影响。方法大鼠随机分为7组:注射用水组、氯诺昔康组、手术致痛组及氯诺昔康预处理组4组(分别于致痛前20min腹腔注射氯诺昔康O.33,1,3,9mg/kg)。3h后取出脊髓,免疫组织化学检测Fos免疫反应阳性神经元(FLIN),观察Fos蛋白的表达。结果手术致痛组脊髓背角浅层Ⅰ~Ⅱ层Fos蛋白大量表达,氯诺昔康可减少Fos蛋白表达,并呈量效关系。结论氯诺昔康对手术致痛引起的脊髓背角Fos蛋白的表达能产生抑制作用,且呈剂量依赖性,同时对手术致痛有镇痛作用。Objective To investigate effects of lornoxicam on c-fos expression of spinal dorsal horn in the rat incision model. Methods 50 rats were randomly divided into seven groups. The first two of which were intraperitoneal injected double distilled water and lornoxicam 3 mg/kg respectively, and the third was intraperitoneal injected double distilled water 20 min prior to the incision in the plantar of the right hind paw. The other four groups also received incision at the same region, and received lornoxicam (0.33, 1, 3, 9 mg/kg) 20 min prior to the incisions. Fos-like immunoreactivity neurons(FLINS) were identified by immunohistochemical technique. Results FLIN were mainly located in the spinal dorsal horn laminae Ⅰ-Ⅱ in those incision groups. The highest density of FLINs was found in the incision group without pretreatment with lornoxicam. Lornoxicam treatment markedly decreased the numbers of FLIN of dorsal horn in a dose-dependent manner. Conclusion Pretreatment with lornoxicam can produce dose-dependent inhibitory effect on c-fos expression in spinal cord dorsal horn. Lornoxicam may reduce post-operative pain.
