摘  要:目的观察切除颌下腺对大鼠睾丸Bax和Bcl 2 mRNA表达的影响。方法切除大鼠颌下腺,分别于术后14、28和42 d处死大鼠,提取睾丸总RNA,反转录,设计特异性引物和Taqman探针,荧光定量聚合酶链反应(PCR)检测Bax和Bcl 2 mRNA的改变。结果随着颌下腺切除时间的延长,实验组大鼠睾丸Bax和Bcl 2 mRNA的比值显著升高,与对照组比较,有显著性差异(P<0.01)。结论颌下腺切除大鼠睾丸Bax和Bcl 2 mRNA比值明显升高,提示颌下腺切除所致的大鼠睾丸生精细胞的凋亡可能是由Bax和Bcl 2介导的线粒体途径进行,但Fas/FasL是否参与该凋亡过程还有待进一步研究。Objective: To detect the changes of Bax and Bcl 2 mRNA expression in rat testis after the removal of submandibular glands. Methods: On day 14, 28 and 42 after operation (bilateral submandibular glands removed or shamoperated), the levels of Bax and Bcl 2 mRNA in rat testis were examined by using fluorogenic quantitative RT-PCR. Results: Fluorogenic quantitative PCR showed that there was a significant increase in the ratio of Bax/Bcl 2 mRNA in rat testis on day 14, 28 and 42 after submanibular glands removed (P〈0.01). Conclusions: The removal of submandibular glands altered the expression of Bax and Bcl 2 mRNA in rat testis and the resulted germ cell apoptosis of rat testis could be related to the mitochondria death pathway mediated by Bax and Bcl 2. The exact mechanism should be further explored.
