摘  要:目的应用表达乙型肝炎病毒(HBV)核心基因(c)与小鼠γ-干扰素(IFN-γ)基因产物的两种重组质粒联合免疫BALB/c小鼠,探讨联合使用表达细胞因子基因重组质粒对DNA免疫的影响及应用价值.方法将HBVc基因与IFN-γ基因重组质粒直接注射到BABL/c小鼠四头肌内,分析免疫后小鼠T细胞增殖、NK细胞杀伤活性及细胞因子释放等免疫应答效应.结果联合γ-IFN基因与HBVc基因免疫小鼠,对T细胞的增殖,NK细胞的杀伤活性及Th1细胞因子的产生等均有显著的增强效应.结论HBVc基因联合γ-IFN基因免疫BALB/c小鼠后,较之单独应用HBVc基因免疫可显著提高抗病毒免疫应答效应,值得进一步开展临床前应用研究.Objective Using hepatitis B viral core gene and interferon-gamma (IFN-γ) gene recombinant plasmid co-combine immunize BALB/c mouse to explove the application values of the cytokine expressing plasmids-mediated immune response. Methods Through direct inject recombinant plasmids DNA into quadriceps muscle of BALB/c mouse, to analysis immune effective including T lymphocytes proliferation, killing activity of natural killer cells and cytokines release. Results IFN- γ expressing plasmids enhance the T lymphocytes proliferation, killing activity of natural killer cells and IFN-γ release than only alone immune with hepatitis viral core gene expressing plasmid significantly. Conclusions After co-combination with IFN-γ expressing plasmid, the immune response which induced by hepatitis B core gene recombinant plasmid augmented obviously. Application of IFN- γ expressing plasmid deserves to undertake pre-clinical explore.
