摘 要:目的观察复方昆明山海棠颗粒剂(FuFangKunmingShanhaitangKeliji,FKSK)主要药效以外的其它药理作用,研究其对整体动物的神经精神系统,心血管系统和呼吸系统的影响,及肝肾功的急性毒副作用。方法给大鼠分别灌胃给予0.5g/kg、1.0g/kg、2.0/kg体重的FKSK后对动物行为活动、饮食、大小便、被毛、皮肤黏膜等进行观察;大鼠麻醉后给药(方式方法及剂量同前),隔一定时间检测其脑电图(EEG)、心电图(ECG)、血压、心率以及呼吸的频率、幅度和张力变化,及取血作肝肾功检查。结果FKSK高、中、低剂量组对大鼠的行为表现、活动姿势、被毛、饮食、大小便、皮肤黏膜、瞳孔均未见异常,对麻醉大鼠的脑电活动亦无明显影响,对麻醉大鼠有轻度减慢心率作用外,对大鼠心电图、血压无明显影响,动物清醒后,减慢心律作用不明显,轻度减慢麻醉大鼠的呼吸频率,降低呼吸幅度和呼吸肌张力,动物清醒后,此作用不明显;对肝功(ALT)肾功(BUN)未见明显影响(P>0.05)。结论FKSK对实验大鼠的精神和神经系统无明显影响;除对麻醉动物有轻度减慢心率和对呼吸肌张力轻度降低外,对心血管系统无明显影响;对大鼠肝、肾功能亦未见急性损害作用。Object To provide laboratory groundwork data of FuFangKunmingShanhaitangKeliji (FKSK) focusing on the therapeutic effects other than the well - known main effects, and investigate its effects on nervous system, cardiovascular system, respiratory system and the acute toxicity and Side effects on hepatic and renal function in animals Methods The animal model of Kunming rats was fed on FKSK and randomly divided into 3 groups. Rats were given FKSK of 0.5,1.0, and2.0 g/( kg · d) in the experimental groups respectively. Different physical parameters such as activities, intake, output, hair, mucocutaneous index were taken, and the rats were fed anesthetically (same method and dose as mentioned above) and were taken EEG,ECG,BP,HR and the respiratory change of rate, extent and tension, as well as blood test of hepatic and renal function. Results FKSK of the high, moderate, low dose group has no effect on the rats' activities, postures, intake,output,hair, mucocutaneous and pupil index; and there was no significant difference in the EEG, ECG and BP results of anesthetic rats, but bradycardiac was observed , as well as depressed respiratory rate, extent and tension, which were mild, but the difference can hardly be observed when the rats were awake. The hepatic and renal function was not significantly effected( P 〉 0.05 ). Conclusion FKSK has no evident pharmacological side-effect on the rats'nervous and mental system. There is not side effect on cardiovascular system, hepatic and renal function of anesthetic rats, except for mild bradycardia effects and respiratory tension depressing.
