环孢素软胶囊在健康人体生物等效性研究

摘　　要：目的:考察北京双鹭药业股份有限公司与丽珠集团丽珠制药厂所研制的环孢素软胶囊的相对生物利用度,评价两者的生物等效性。方法:18名健康男性志愿者单剂量口服试验制剂或参比制剂,高效液相色谱(HPLC)测定全血中药物浓度,以内标法计算血药浓度。结果:受试制剂及参比制剂环孢素实测平均血药峰浓度Cmax分别为(1144.87±382.38)和(1241.08±383.31)μg·L-1;实测平均达峰时间tmax分别为(2.14±0.61)和(1.83±0.59)h;受试制剂及参比制剂t1/2(ke)分别为(3.54±0.92)和(4.41±2.51)h;AUC0-tn平均值分别为(4698.15±1872.31)和(4873.95±1805.63)!g·L-1·h;AUC0-∞平均值分别为(5564.81±2059.53)和(6157.76±2314.98)!g·L-1·h;试验制剂环孢素的相对生物利用度F0-tn、F0-∞分别为(98.99±20.40)%和(95.37±25.7)%。结论:受试制剂和参比制剂具有生物等效性。Objective： To evaluate the bioequivalence of cyclosporine soft capsule of test and reference. Methods： The whole blood concentrations of 18 male healthy volunteers after a single oral dose of cyclosporinc soft capsule of test and reference preparations were determined by HPLC method. Results： Cmax,tmax,t1/2（ke）,AUC0-∞ of test preparation and reference preparation were （1 144.87 ± 382.38） and （1 241.08 ± 383,31 ）μg·L^-1; （2.14±0,61 ） and （1.83 ±0.59） h; （3,54 ±0,92） and （4,41 ± 2,51 ） h; （4 698.15 ± 1 872.31） and （4 873,95 ± 1 805.63） μg·L^-1·h; （5 564.81 ± 2059,53） and （6 157,76 ±2314.98） μg·L^-1·h respectively, and F0-tn, F0-∞ of test preparation were （98,99 ± 20.40）% and （95.37 ± 25.7）%. Conclusion： The test preparation was bioequivalcnt to the rcference preparation
