三虎丸对佐剂性关节炎大鼠腹腔巨噬细胞产生TNFI-α和IL-1的影响  被引量:2

摘  要:目的探讨三虎丸治疗类风湿性关节炎的免疫调节机制。方法将SD大鼠24只制成佐剂性关节炎(AA)模型,分为三虎丸组、益肾躅痹丸组、模型对照组、空白对照组,造模第18d开始,治疗组和益肾躅痹丸组分别灌服三虎丸和益肾蠲痹丸,模型对照组、空白对照组灌服蒸馏水。于给药后1w腹腔注射30g/L硫代羟基乙酸盐2mL/只诱导腹腔产生巨噬细胞(M)φ,于第4天给药2h后断颈放血处死,收集血清免疫复合物(CIC)、总补体(CH50);收集腹腔Mφ检测白细胞介素(IL)-1、肿瘤坏死因子(TNF)α活性。结果三虎丸能明显降低CH50,与模型对照组比较有显著性差异(P<0.01),与益肾躅痹丸组比较无显著性差异(P>0.05);在降低AA大鼠血清CIC含量方面,三虎丸组与益肾躅痹丸组比较具有显著性意义(P<0.01);三虎丸在抑制脂多糖(LPS)诱导腹腔Mφ产生IL-1活性方面,与益肾躅痹丸组比较具有显著性差异(P<0.01),在抑制LPS诱导腹腔Mφ产生TNFα活性方面,明显优于益肾蠲痹丸组(P<0.05)。结论三虎丸能降低AA大鼠血清CH50、CIC含量,抑制LPS诱导AA大鼠腹腔Mφ产生IL-4、TNFα的活性,具有良好的免疫调节作用。Objective To study the immune modulatory effect of Sanhu Tablet(ST) on treatment of rheumatoid arthritis. Methods The adjuvant arthritis(AA) rat models were divided randomly into 3 groups as model control group,, ST group and Yishen zhuanbi tablet(YZT) group which were treated with ST and YZT respectively and blank contrast group was also established, which was fed distillation water. After 1 week (26th day),the macrophages(Mφ) in abdominal cavity was induced by ip 30g-IAR 2ml per rat in each group, then, all rats were decapitated at 2nd hour after 4 days of the fed. The artery blood was collected to measure circulating immune complex (CIC) and the total complement(CH50); and the macrophages(Mφ) was collected to also measure the activity of Inter-leukin(IL-1)and tumor necrosis factor α(TNF). Results To decrease the serum concentration of CIC and inhibit the activity of LI-1 and TNFα, 0.05 or P〈0.01 ). Conclusion The immune regulating effect of ST may be one of the mechanism treating rheumatoid arthritis.
