Significance of Expression of Brain-Derived Neurotrophic Factor and Neuron Apoptosis in Brain of Rats with Repeated Febrile Seizures

摘  要:目的探讨大鼠反复高热惊厥(FS)后脑源性神经营养因子(BDNF)的变化。方法雄性SD大鼠51只随机分为健康对照组(NC组,n=14),高热对照组(HC组,n=19)和FS组(n=18),热水浴法建立FS模型。酶联免疫吸附试验(ELISA)测定各组海马匀浆BDNF水平,免疫组织化学法检测BDNF在大鼠脑组织各区的表达,原位末端标记法(TUNEL)检测大鼠脑组织细胞凋亡,采用SPSS13.0软件进行统计学分析。结果ELISA法测得的FS组海马BDNF水平(89.90±12.51)ng/g明显高于NC组(54.43±18.92)ng/g和HC组(64.09±15.03)ng/g(Pa<0.01),FS组大鼠各脑区BDNF阳性神经元A值亦明显高于NC和HC组(Pa<0.01);FS组分析各区凋亡指数(AI)均明显高于NC与HC组(Pa<0.01),各区AI从高到低依次为海马齿状回(DG)区(32.65±2.14)%、CA3区(28.99±1.16)%、CA1区(24.28±0.92)%、颞叶皮层(CTL)(22.19±1.06)%,且原位末端标记法染色观察脑组织各区细胞凋亡程度与BDNF在脑组织各区的表达特点呈正相关(r=0.332P<0.05)。结论反复FS后大鼠脑内BDNF表达明显增加,并且与神经细胞的凋亡程度相关。Objective To explore the changes of brain - derived neurotrophic factor (BDNF) after repeated febrile seizures (FS). Methods Fifty - one male SD rats were divided into 3 groups randomly : normal group ( NC group,n = 14) , hypertherrnic group ( HC group, n = 19) ,FS group(n = 18). FS were induced by hot water bath. Tile level of BDNF in hippocampus homogenate were measured by enzyme linked immunosorbent assay (ELISA) and the expression of BDNF were measured by immunohistochemistry. The apoptosis of the brain cells were determined by terminal deoxynucleotide mediated nick end labeling (TUNEL). The results were analysed with the software of SPSS 13.0. Results The level of BDNF in hippocampus in FS group(89.90 ± 12.51 ) ng/g was significantly higher than that in NC group(54.43 ± 18.92) ng/g and HC group(64.09 ± 15.03 ) ng/g (Pa 〈 0.01 ) ,In FS group, the A score of the BDNF positive neurons in brain region was more than that in NC group ( P 〈 0.01 ) and HC group ( P 〈 0.01 ) ; The apoptotic index (AI) in FS group was significantly highter than that in NC group and HC group (P 〈 0.01 ), and the AI from high to low were dentate gyrus (DG) ( 32.65 ± 2. 14 ) %, CA3 ( 28.99 ± 1.16) % , CA1 (24.28 ± 0.92 )% and cortex of temporal lobe AI (22. 19 ±1.06 )%. The expression of BDNF in brain region and the AI in brain region desmonstrated positive correlation ( r = 0. 332 P 〈 0.05 ). Conclusion Repeated FS can obviously increase the expression of BDNF, and correlate with neuron apoptosis.
