摘 要:报道了通过茎段离体培养快速繁殖苦丁茶树的研究结果:筛选出了比较理想的初代培养基,在该培养基上茎段腋芽的萌发率在80%以上,扩大繁殖阶段腋芽萌发率在90%以上,并有18%-20%的材料形成丛芽;生根培养阶段95%以上的无根苗均能形成良好的根系;试验了有根苗移栽法和无根苗移栽法,两种方法的移栽成活率均达到100%.This paper dealt with the rapid propagation of Kuding tea tree (Ilex latifolia thunb. )via stem segment culture in vitro. The experimental results were shown as follows.1. Of all media adopted in the initial culture, the medium-H Macroelements + MS Microelements + 100ng/L Inositol + 0. 5 mg/L Nicotinic acid + 1 .0mg/L Thiamine. HCI + 0.5mg/L Pryidoxine HCI + 2.0mg/L Glycine + 1.0 L 6-BA + 0.5mg/L KT + 0.5mg/L IBA+ 2.0% Sucrpse +0.7% Agar, was the Best,and the germination late of the axillary buds was more than 80%.2. The medium used in the initial culture, or the medium-MS + 100mg/L Inositol+ 0.5mg/L Nicotinic acid + 1 .0mg/L Thiamine. HCI + 0.5mg/L Pryidoxine HCI + 2. 0mg/L Glycine + 1. 5mg/L 6-BA + 0.5mg/L KT+ 0.5mg/L IBA + 2.0% Sucare + 0.7% Agar,could be used in the proliferation culture.The germination rates of the axillary buds on both media were mord than 90%,and 18% 20% d the explants fond clustered shoots.3. The medium-MS Fe-salt + 1/3 other inorganic nutrients of MS + 100mg/L Inpsotol+1 .0mg/L Nicotinic acid + 2.0mg/L Thiamine. HCl + 1. 0mg/L pryidoxine HCI + 4. 0mg/L Glycine +1 .5% Sucrose + 1.0% Agar+ 1 .5% Active carbon + 2.0mg/L ABT-1 rooting powder, was more suitable for the induction of the root emergence of Kuding tea shoots, and more than 95% formed good rootages.4. Both shoots and complete planilets were used in the transplanting tests, and the survival pereentages of them were 100%.
