摘  要:研究了钼(Ⅵ)-水杨基荧光酮(SAF)配合物作为光谱探针与蛋白质相互作用及光谱性质。实验结果表明,在pH=1.00~2.50的HCl-NaAc缓冲介质中,在乳化剂Tween-40存在下,蛋白质与Mo(Ⅵ)-SAF配合物发生作用,导致配合物溶液褪色,其最大吸收波长处吸光度的降低值与蛋白质的浓度成正比。基于此建立了以Mo(Ⅵ)-SAF配合物为光谱探针,分光光度测定蛋白质含量的新方法。该法灵敏度高,选择性好。蛋白质(以牛血清白蛋白为例)的浓度在0 mg/L^12 mg/L范围内服从比尔定律,表观摩尔吸光系数ε579=2.85×106L.mol-1.cm-1。生物体内常见物质均不产生干扰,直接用于人尿,新生牛血清的测定,回收率分别为98.8%和101.5%。The interaction and spectral characters of Salicylfluorone-Molybdenum(Ⅵ) and protein were studied. In the buffer meditun of HC1-NaAe at pH 1.80 and in the presence of Tween-40, after being heated for 20rain in the temperature of 60 ℃, SAF-Mo( Ⅵ) complex interaet with protein to form a stable compound, the absorption peak value was at 578.8 nm. Beer's law was obeyed in the range of 0 ~ 12 nag , L^-1 of protein. The molar absorptivity of complex at 578.8 nm was 2.85×10^6 L/( tool , era). The method possessed high sensitivity as well as high selectivity, h could be applied to assay protein in human urine and bovine serum, the recovery was 98.8% and 101.5% respectively.
