摘 要:[目的]探讨骨髓间充质干细胞(MSCs)的免疫负调节作用。[方法]分离和鉴定大鼠MSCs,用MTS法观察该细胞对淋巴细胞增殖活性的影响,乳酸脱氢酶(LDH)释放法检测MSCs与淋巴母细胞混合培养液24h后上清中LDH活性。[结果]大鼠MSCs表型为CD29+、CD44+、CD45-、CD54+、CD95+和SH-2+,具有诱导分化成脂肪和成骨的潜能力,对ConA刺激的淋巴细胞增殖有显著的抑制作用,能够诱导淋巴母细胞裂解,释放LDH。[结论]MSCs能够抑制淋巴细胞的活化增殖,这种负向免疫调节功能有可能在免疫耐受诱中发挥作用。[Objective] To explore the inhibitory effect of negative inmnmoregulatory activity of mesenchymal stem cells (MSCs). [ Methods] Tile MSCs were identified and isolated from SD rat. The effect of MSCs on the lymphocyte proliferation was observed by MTS. The activity of LDH in supematant of the mixed culture medium of MSCs and lymphoblast for 24 hours were detected by LDH releasing method. [Results] The phenotypes of MSCs ill rats were CD29^+, CD44^+, CD45^+, CD54^+, CD95^+ and SH-2^+, which had potential capability to induct differentiation to be fat and bone cells, obviously inhibited the lymphocyte proliferation stimulated by ConA, and induce the lymphoblast disruption to release LDH. [Conclusion] MSCs could inhibit the activity mid proliferation of lymphocytic, the negative immunorgulatory of which could showed an obviously effect on the inununologic tolerance.
