摘  要:目的建立同时测定舒胆胶囊中大黄酸、大黄素、大黄酚、大黄素甲醚含量的方法,完善现有质量标准。方法采用HPLC法。用Diamonsil C18柱(200mm×4.6mm,5μm)分离,用甲醇-水-磷酸(体积比84:16:0.1)进行等度洗脱,流速为1.0mL·min^-1,柱温为35℃,检测波长为254nm。结果大黄酸质量浓度在0.2200~4.400mg·L^-1内、大黄素质量浓度在0.1620~3.240mg·L^-1内、大黄酚质量浓度在0.5208~10.42mg·L^-1内、大黄素甲醚质量浓度在0.1092~2.184mg·L^-1内与峰面积呈良好的线性关系,相关系数分别为0.9997、0.9999、0.9998、0.9999;平均回收率分别为100.5%(RSD=0.84%)、100.4%(RSD=0.51%)、100.8%(RSD=0.29%)、99.8%(RSD=0.47%)。结论本方法简便、快捷,结果准确,重复性好,可作为舒胆胶囊的质量控制方法之一。Objective To establish an HPLC method for simultaneous determination of rhein, emodin, chrysophanol and physcion in Shudan (traditional Chinese medicines)capsules. Methods The chromatographic method was applied on a Diamonsil C18 column (200 mm × 4.6 mm, 5 μm) with methanol-water-phosphoric acid ( V : V : V = 84 : 16 : 0.1 ) as the mobile phase. The flow rate was 1.0 mL· min^- 1 and column temperature was 35 ℃. The detection wavelength was set at 254 nm. Results The calibration curve was linear within the range of 0. 220 0 - 4. 400 mg·L^-1 ( r = 0. 999 7) for rhein, 0. 162 0 - 3. 240 mg·L^-1 ( r = 0. 999 9) for emodin, 0.520 8 - 10.42 mg·L^-1(r = 0.999 8) for chrysophanol, 0.109 2 - 2.184 mg·L^-1(r = 0. 999 9) for physcion ; the average recoveries were 100.5 % (RSD = 0.84 %) for rhein, 100.4 % (RSD = 0.51% ) for emodin, 100.8 % (RSD = 0.29 % ) for chrysophanol, 99.8 % (RSD = 0.47 % ) for physcion. Conclusions The method is simple, accurate and reproducible and can be used for the quality control of Shudan capsules.
