脂多糖对大鼠中性粒细胞基质金属蛋白酶-8 mRNA表达的影响

摘　　要：目的:观察脂多糖(LPS)对体外培养大鼠中性粒细胞表达MMP-8 mRNA的影响。方法:抽取大鼠外周血,分离培养中性粒细胞,随机分为5组,前4组(实验组)加入1mg/L LPS分别孵育30min、60min、90min和120min,第5组加入等体积1mg/L PBS作为对照组,采用RT-PCR法检测MMP-8 mRNA的表达。结果:对照组中MMP-8 mRNA的表达水平较低,LPS刺激后各组表达均升高(P<0.05或0.01),90min组MMP-8 mRNA的表达水平最高。结论:LPS可以刺激大鼠中性粒细胞表达MMP-8 mRNA。Aim： To observe the influence of LPS on the expression MMP-8 mRNA in cultured rat neutrophits. Methods ：Neutrophils from SD rats were isolated and then allocated into five groups：the first 4 groups （LPS groups） were added 1 mg/L LPS and cultured for 30 min, 60 min, 90 min and 120 min,respectively. The 5th group（control group） was given 1 mg/L PBS. MMP-8 mRNA was measured by RT-PCR. Results：There was lower expression level of MMP-8 mRNA in control group compared with LPS groups （P 〈 0.05 or 0.01 ）. The expression level of MMP-8 mRNA reached peak in 90 min group. Conclusion ： LPS can induce the expression of MMP-8 mRNA in neutrophils.
