甲基莲心碱增强伊马替尼、多柔比星对K562/G01细胞敏感性的研究

摘　　要：目的探讨甲基莲心碱(Nef)在耐伊马替尼多药耐药细胞株(K562/G01)对STI571、多柔比星敏感作用的影响。方法采用改良MTT法观察STI571或DOX单用与联用Nef对K562/G01细胞增殖抑制的变化。结果STI571(20μmol·L-1)或DOX(1μmol·L-1)单用细胞抑制率(37.72±2.17)、(29.51±2.28);与Nef(8μmol·L-1)联用细胞抑制率明显升高(P<0.01),分别为(88.64±3.04)、(51.93±3.52),逆转倍数分别为(2.35±0.04)、(1.76±0.02)。结论Nef能增强K562/G01细胞对STI571、DOX的敏感性。Objective To explore the effect of neferine on the sensitivity of imatinib（STI 571）and doxorubicin（DOX）to K562 /G01 cells.Methods Improved MTT assay was used to observe the alteration of the proliferation of K562 /G01 cell line treated with STI 571 or DOX alone or combinated with neferine.Results The inhibiting rates on K562/G01 cells of STI 571（20 μmol·L^-1）or DOX（1 μmol·L^-1）were（37.72±2.17）and（29.51±2.28）.When combined with neferine（8 μmol·L^-1）,their inhibiting rates were increased to（88.64±3.04）and（51.93±3.52）,which were significanly higher than those of STI 571（20 μmol·L^-1）or DOX（1 μmol·L^-1）（P〈0.01）,and their reverse fold was（2.35±0.04）and（1.76±0.02）,respectively.Conclusion Neferine significantly enhances the sensitivity of STI 571 and DOX to K562 /G01 cells.
