摘  要:通过对鞍带石斑鱼Epinephelus lanceolatus、棕点石斑鱼E.fuscoguttatus、斜带石斑鱼E.coioides、鲑点石斑鱼E.fario的线粒体细胞色素b基因全序列扩增、克隆及测序,得到1141bp序列,并结合GenBank其他7种石斑鱼同源序列进行比较分析,共发现429个核苷酸位点存在变异(37.6%)。序列中的转换大于颠换,碱基替换多发生于密码子第3位。以4种鲈形目鱼类为外群,用MEGA3.0软件中的NJ、ME、MP等3种方法构建分子进化树,结果显示,鞍带石斑鱼与石斑鱼属鱼类有较近的亲缘关系,支持鞍带石斑鱼归入石斑鱼属。The mitochondrial cytochrome b gene of groupers, Epinephelus lanceolatus, E. fuscoguttatus, E. coioides, and E. fario, were amplified, cloned and sequenced. The 1 141 bp sequence of cytochrome b gene in four groupers were obtained. Compared with the cytochrome b gene sequence of the other seven groupers from the GenBank, 429 nucleotide sites were varied (37. 6%) among 1141 bp. The number of transitions is more than that of transversions; most substitutions occurred in the third position of the co- don. With four species of Perciformes designated as outgroups, the molecular phylogenetic trees constructed by NJ, ME, MP methods of MEGA3.0 indicates that E. lanceolatus had a closer relationship with Epinephelus, suggesting it may be reasonable that E. lanceolatus belongs to genus Epinephelus.
