摘  要:目的探讨线粒体DNA(mtDNA)5178A/C,10398A/G多态性与中国新疆维吾尔族长寿老人的关联性。方法采用聚合酶链反应-限制性片段长度多态性(PCR—RFLP)的分析方法,将165例新疆维吾尔族长寿老人分为百岁组(≥100岁)65例,长寿组(91~99岁)100例和112例对照组(≤70岁)比较进行研究。结果在百岁组中mtDNA 5178A等位基因频率高于对照组(P=0.0029),而百岁组和长寿组10398G等位基因的频率显著高于对照组(P〈0.005)。百岁组和长寿组中mtDNA 5178A/10398G基因型(A—G单倍型)组合明显高于对照组。结论线粒体DNA 5178A和10398G等位基因与新疆维吾尔族长寿相关。5178A和10398G等位基因及A—G基因型组合(A—G单倍型)为长寿的保护因素。Objective Sequence variations in mitochondrial DNA genes have been found to influence successful aging and longevity. The aim of this study was to examine whether mitochondrial DNA polymorphisms influenced the longevity in Chinese Uygur population. Methods We genotyped the mt5178A and mt10398G in 277 sampels representing three age group (centenarian, n = 65 ; longevity, n = 100; controls, n = 112) from Taklimakan desert Uygur centenarians,China Results The distribution frequency of the 5178A in the centenarian ( 51/65, 78.5 % ) is slightly higher than in those of controls ( 63/112,56.3 % , P = 0. 0029 ) . The frequency of the 10398 G were significantly higher among the centenarian(47/65,72.3% )and longevity(68/100,68% ) in comparison to the controls(60/112,53.6% , P 〈 0. 005 ). Analysis on 2 - polymorphism combinations showed that the frequency of A - G genotypes combination of 5178/10398 ( A - G haplotype) was significantly higher in the centenarian and longevity groups than that in the controls. Conclusion These results suggest that 5178A and 10398G genotype of mitochondrial ND2 gene polymorphisms are contribution to the longevity. Furthermore, A - G genotypes combination ( A - G haplotype) of 5178/10398 polymorphisms were the advantegeous factor of longevity, resulted from long - term genetic isolation in Uygur in Xinjiang Hetian.
