摘  要:目的研究细薄星芒海绵Stelletta tenuis Lindgren中总三萜提取物的高效液相色谱指纹图谱,为控制和评价其质量提供可靠方法。方法采用HPLC-PAD方法,梯度洗脱,色谱条件为YMCPack—SIL分析柱(250mm×4.6mm,5μm);流动相A为正己烷,流动相B为乙酸乙酯,梯度洗脱,分析时间50min,0~30minA(75%),30~35minA(75%~3%),35~50minA(3%);流速1.0mL·min^-1;检测波长:360nm;柱温:室温;进样量:20μL。结果从10批海绵总三萜提取物样品指纹图谱中可以发现7个共有峰,其中保留时间在0~30min有4个特征指纹峰,分别为峰1,2,3和7,通过与对照品的保留时间和紫外光谱信息比较,可以确定它们分别为(1)Stellettin D;(2)Stellettin A;(3)Stellettin B;(7)Stellettin C。不同批次细薄星芒海绵总三萜提取物的HPLC图谱之间具有很好的相关性。结论本研究建立的细薄星芒海绵Stelletta tenuis Lindgren总三萜提取物的高效液相色谱指纹图谱的特征性和专属性强,可结合特征指纹峰的含量测定有效控制样品的质量。Objective To establish a sensitive and specific HPLC fingerprint method for controlling and evaluating the quality of total isomalabaricanes from Stelletta tenuis Lindgren. Methods HPLC-PAD method was applied for quality and quantitative assessment of the pharmaceutical extracts from Stelletta tenuis Lindgren. HPLC analysis was performed on a YMC Pack-SIL column (250 min×4.6mm, 5μm) with n-hexane and ethyl acetate as mobile phase and the flow rate was 1.0mL · min^-1. Results The HPLC chromatographic fingerprint of the total isomalabaricanes, showing 7 characteristic peaks and four main peaks in 0~30 min of retention time was obtained from 10 batch of samples. By comparison of the retention time and on-line UV spectra of chemical standards, peak 1, 2, :3 and 7 were identified as(1)Stellettin D; (2)Stellettin A; (3)Stellettin B; (7)Stellettin C, respectively. Moreover, comparison of the HPLC profiles of the total isomalabaricanes, the extract prepared indicated that they were closely related to each other. Conclusion The HPLC profiles and quantitative assessment of the total isomalabaricanes from Stelletta tenuis Lindgren with high characterization and specificity can be used to effectively control their quality .
