摘 要:选用10个水稻基因型诱导愈伤组织,探讨在固体与液体培养条件下愈伤组织的增殖情况。结果表明,水稻愈伤组织诱导率与固体继代培养增殖在基因型间存在差异,徐稻4和9823的愈伤组织诱导率和固体继代增殖效果都较好。影响液体培养的主要因素依次是基因型、6-BA浓度、2,4-D浓度和KT浓度。在发酵罐中愈伤组织培养随着培养时间的增加,增量也增加,96h后增长量趋于稳定。水稻有可能用作植物生物反应器。The callus was induced from mature embryos of ten genotypes in rice, and then the proliferation in solid and liquid culture was studied in our experiment. The results showed that there were differences in induction rate and proliferation profile among those genotypes in solid culture. Compared with others, the Xudao4 and 9823 produced the highest induction rates and callus proliferations. While in liquid culture process, the dominant factors determining the proliferation was the genotype itself, plus the influence of the concentrations of 6 BA, 2,4-D,and KT with gradually weakening effect. In the fermentation environment by contrast, the callus grew further with time and eventually reached saturation by the time of 96 hours. Our research clearly showed that the rice could be considered as a good candidate for plant bioreactor.
