肝包虫新鲜组织标本库的初步建立及鉴定

摘　　要：目的：探讨初步建立包虫新鲜组织标本库的条件，摸索一套适合我院实际情况的建库方法，为更加系统开展进一步的相关研究做好准备。方法：选取2004年7月～2007年8月期间在我院肝胆外科以肝包虫病为主要诊断而行手术切除的患者的手术标本，分割后用编号的灭菌锡箔纸包好，迅速投入液氮转移罐，然后移至深低温冰箱长期保存。全部病例均经过临床与病理学检查予以证实。随即抽取其中部分组织使用Trizol试剂提取总RNA，以鉴定组织成分的完整性。结果：到目前为止共采集标本120例，随机抽取其中11份标本，经Trizol试剂提取总RNA，经1％琼脂糖凝胶电泳检测未见显著降解，经紫外分光光度计分析得出其OD360／OD280=1．6～2．0。结论：本实验所采用的标本采集方法可保证标本库的质量，有利于更有效地利用病例资源。Objective： To explore a rational and effective operative procedure of fresh tissue samples preservation for hepatic hydatid,and to establish a tissue bank with high quality,the authors had made a plan to store up the tissue samples of hepatic hydatid resected during operaion in deeplow temperature, which is based on the clinical preponderance in the treatment of this diseases of our hospital,and will be a good foundation for the systemic basic research of hepatic hydatid.Methods： Cases of hepatic hydatid confirmed by pathology were selected from the inpatients of Department of Hepatobiliary Surgery in our hospital from Jul.2004 to Aug. 2007 . Fresh tissues were taken from the excised sample. They were stored in liguid nitrogon container temporarily, and transferred into refrigerator for longtime storage as quickly as possible. Pathological diagnosis must be made on paraffin embedded samples. A part of the tissues was used to isolate total RNA by Trizol reagent for integrality judgment of it.Results：There is 120 excised samples were stored as yet,and select 11.The precipitated total RNA could be seen at the bottom of Eppendorf tube, of which the rate of OD260/OD280 was calculated about 1.6 to 2.0.It also could be seen in the 1% agarose gel electrophoresis for the obvious two bands of.28s and 18s. Conclusion：The present program for hepatic hydatid tissue severance and storage is feasible and could supply the good - quality sample for further study.
