摘 要:目的探讨肾上腺髓质素受体(ADMR)在动脉粥样硬化(AS)大鼠胸主动脉病变时的变化及氟伐他汀对ADMR表达的影响。方法将SD大鼠随机分为3组:正常对照组、AS模型组和氟伐他汀干预组。采用喂养高脂饲料(正常饲料基础上加入30g/kg胆固醇、5g/kg胆酸钠、2g/kg丙基硫氧嘧啶、50g/kg精制糖、100g/kg猪油)加维生素D3制备AS动物模型,氟伐他汀20mg/(kg·d)灌胃12周。用RT-PCR和Western印记法分别检测各组胸主动脉中ADMR mRNA和蛋白的表达情况。结果氟伐他汀干预组大鼠与AS组比较,体质量差异无统计学意义;与AS组相比,干预组三酰甘油(TG)、总胆固醇(CHOL)显著下降(P<0.01);与正常对照组相比,模型组TG、CHOL明显升高(P<0.05);与正常对照组相比,模型组、干预组血钙均明显升高(P<0.05)。氟伐他汀干预组大鼠胸主动脉中ADMR mRNA及蛋白的表达均显著高于AS组(P<0.05)。结论氟伐他汀可上调AS大鼠胸主动脉ADMR基因和蛋白的表达,ADMR可能参与了氟伐他汀抗动脉粥样硬化的作用。AIM To investigate the changes of ADMR (adrenomedullin receptor) in ascending aorta of AS rats and the effects of fluvastatin on the expression of ADMR mRNA and protein. METHODS Fortyfive male SD rats were randomly divided into 3 groups: normal control group (N), atherosclerosis group (AS) and atherosclerosis rats treated with fluvastatin (F). Group AS were fed with high lipid feedstuff (including 30 g/L cholesterol, 5 g/kg cholic acid natrium, 2 g/kg propyl thiouracil, 50 g/kg refined sugar, 100 g/kg lard oil) plus vitamin D3, and group F were fed as group AS, followed with fluvastatin (20 mg/kg/d, intragastric administration). The mRNA and protein expressions of ADMR in ascending aorta of rats were assessed by reverse transcription PCR and Western blot respectively. RESULTS ①The values of body weight showed no significant difference between group F and group AS. The triglyceride and cholesterol levels in group F were reduced significantly compared with those in group AS ( P 〈 0. 01 ) and the levels in group AS were increased significantly compared with those in group N (P 〈 0. 05 ). The blood serum calculus levels in group AS and group F increased significantly compared with those in group N (P 〈 0. 05). ②The ADMR mRNA level and the expressions of ADMR protein in the ascending aorta in group F increased compared with those in group AS ( both P 〈 0. 05 ). CONCLUSION Fluvastatin treatment improves the expressions of ADMR mRNA and protein in the ascending aorta and the ADMR may play an important role in anti-atherosclerosis of fluvastatin.
