摘 要:目的探讨肿瘤坏死因子-α(TNF-α)及核因子-κB(NF-κB)在铍病发病中的作用。方法采用一次性非暴露式气管注入法染毒建立铍病模型。用酶联免疫吸附法(ELISA)检测肺组织匀浆及支气管肺泡灌洗液(BALF)中TNF-α水平,免疫组化法测定肺组织NF-κB的表达。同时HE染色观察肺组织的大体病理改变。结果大鼠模型组肺组织的病理改变基本符合人类铍病的特点。模型组大鼠肺组织匀浆及BALF中TNF-α水平、肺组织中NF-κB的表达较生理盐水对照组明显升高(P<0·01);且模型组内TNF-α的水平、NF-κB的表达,60d组明显高于20d组(P<0·05)。结论TNF-α和NF-κB相互作用,在铍病的发病机理中起着重要作用,并可作为判定疾病活动性的一项重要参考指标。Objective To explore the role of TNF-α and NF-kB played in the pathogenesis of experimental berylliosis in rats. Methods The rats were intratracheally injected with 10mg/ml beryllium oxide (BeO) 0. 5ml, then observing the histopatho-logical changes of lung, the TNF-α level ( with ELISA ) and the expression of NF-kB ( by immunohistochemistry method) in lung tissues and BALF of the rats, 20 d, 40 d and 60 d after the injection respectively. Results The results showed that the pathological changes were consistent with the basic features of berylliosis; both the TNF-α level and the expression of NF-kB in lung tissues and BALF were all significantly risen in berylliosis rats compared with the control rats (P 〈0. 01 ) ; and all the results mentioned above in 60 d group were significantly higher than that of 20 d group (P 〈 0. 05 ). Conclusion TNF-α and NF-kB might play some important role in the pathogenesis of berylliosis, which might be the reference indicators of developing degree of the disease.
