鲁西斗鸡IGF-Ⅰ基因多态性与体型指标关系的研究

摘　　要：采用PCR-RFLP技术,对124只鲁西斗鸡IGF-Ⅰ基因的5′端调控区进行遗传多态性研究,并初步探讨其基因型与体型指标的相关性。将扩增产物进行PstⅠ酶切后出现AA、AB和BB3种基因型,其基因型频率分别是0.4516、0.3710、0.1774,等位基因A、B的频率分别是0.637、0.363。对纯合子进行测序,发现在IGF-Ⅰ基因5′端距启动子7.8kb处有A→G的转换。经χ2检验,鲁西斗鸡在该位点上处于Hardy-Weinberg平衡状态(P﹥0.05)。分析基因型与16周龄体重体尺时发现:在体重、胸宽、胸骨长指数和胫长指数方面,存在BB>AB>AA的趋势,但在颈长和腿长方面,存在着AA>AB>BB的趋势。表明"A"等位基因与颈长和腿长相关,含此基因个体适合向着斗性强的方向选育,"B"等位基因与体重、胸宽、胸骨长和胫长指数相关,含此基因个体适合向肉用方向选育。The research on the genetic polymorphism of the IGF-I gene＇s 5＇ region in 124 Luxi game chicken was did by PCR-RELP,the correlation between the genotype and somatotype index was analyzed. PCR products appeared AA,AB,BB genotypes treated by Pst I ,which frequency was 36.88%,49.65%,13.48%,respectively. The frequency of A and B was 0.637 and 0.363,respectively. The transition A to G was detected at 5＇ region of IGF-I gene where was 7.8 kb from promoter by sequencing the homozygoles. Checked by X^2,Luxi game chicken was placed on Hardy-Weinberg balanced state （P〉 0.05） on the site of IGF-I. Genotype,body weight and size of 16 weeks age was analyzed. The results showed that the tendency of BB〉AB〉AA was found in body weight,chest width,breastbone index,shank index,but the tendency of AA〉AB〉BB was found in neck length and shank length. In conclusion, ＂A＂gene was relative to shank length and neck length,could be used to breed stronger belligerence type. ＂B＂gene was relative to body weight,chest width,breastbone index and shank index,could be used to breed meat-producing type.
