摘 要:目的探讨促心房纤维化因素转化生长因子β1(TGF-β1)和血管紧张素转化酶(ACE)与心房颤动(AF)的相关性。方法健康杂种犬16条,随机分为对照组(n=8)与AF组(n=8)。AF组置入永久起搏器,起搏时间24周。对照组不植入起搏器。制备AF犬模型。在左、右心房切取组织块,甲醛固定后,石蜡包埋保存。两组石蜡切片均行免疫组织化学检测TGF-β1和ACE,比较两组间TGF-β1、ACE的表达水平。结果以平均光密度表示各组TGF-β1、ACE的表达水平,进行定量分析。结果显示:AF组TGF-β1的平均光密度测定值明显高于对照组(19.00±6.44)%vs(11.07±6.27)%(P<0.01);AF组ACE的平均光密度测定值明显高于对照组(8.73±3.86)%vs(5.52±2.66)%(P<0.01)。结论TGF-β1和ACE是促进心房纤维化形成的重要因素,可能与AF的发生和维持密切相关。Objective To investigate whether atrial expression of transforming growth factor β1 (TGF-β1) and angiotensin-convertion enzyme(ACE) is altered in dogs with atrial fibrillation(AF). Methods Sixteen mongrel dogs were randomly divided into two groups:paclng group( n=8) and control group( n=8). High frequency pacing (500±20) times/min was performed in pacing group with permanent pacemaker. Pacing was not performed in control group. After pacing to 24 weeks,tissue samples were obtained from the left and right atria when dogs were killed. The level of TGF-β1 and ACE in atria were detected by immunohistochemistry. Results Compared to those of control group, the expression levels of TGF-β1 and ACE were significantly enhanced in atrial tissues during AF in pacing group, (19.00±6.44)% vs (11.07±6.27)%,(8.73±3.86)% vs (5.52±2.66)%(P〈0.01). Conclusion TGF-β1 and ACEin dog participate in AF, which may be one of the main mechanisms in the genesis and development of AF with pomoting atrium fibrosis.
