摘 要:为给小麦育种提供基础材料,利用SSR技术和高分子量麦谷蛋白SDS-PAGE对由EMS诱导河农822获得的矮秆、短芒、密穗、棒状穗、早熟等8个小麦性状变异株进行鉴定。结果表明:在DNA水平,除矮秆变异株外,其余均缺失了1条50-190 bp的片段,各个不同性状突变株均增加了3-4条44-574 bp的片段;在蛋白质水平,不同性状突变株高分子量麦谷蛋白亚基在Glu-1B和Glu-1D位点呈现出缺失和新增亚基2种情况。这表明突变材料与对照相比,不仅在生物学形态存在明显差异,而且在DNA和蛋白质水平上也产生了突变,为小麦突变产生提供了充分的分子生物学证据。By using SSR molecular marker and high-molecular weight subunits of glutenin SDS- PAGE, the wheat cultivars HeNong822 and 8 character-mutant types obtained from the said cultivars induced by EMS, such as dwarf, awnless, dense panicle, clubbed panicle and early flower, were identified. The results of SSR analysis indicated that other mutant types deleted fragment of 50--190 bp, but all mutant types increase by 3--4 fragments of 44--574 bp except the dwarfing type. The identification of glutenin SDS- PAGE demonstrated that the different character mutations may add/delete a subunit on Glu- 1B and Glu - 1D. Compared with the control cultivars, the mutants varied not only on the biology modality, but also on the DNA and protein level, which provided sufficient evidence of molecular biology for the production of wheat mutant.
