摘  要:目的:探讨中药茵陈蒿对雌孕激素诱导的肝内胆汁淤积孕鼠肝脏超微结构、细胞黏附分子-1(ICAM-1)表达的影响。方法:运用苯甲酸雌二醇和黄体酮诱导妊娠期肝内胆汁淤积症大鼠模型,将60只孕鼠随机分为正常组、模型组、茵陈蒿高、中、低剂量组和熊去氧胆酸组。将各组孕鼠取肝脏组织进行光镜和电镜观察,并运用免疫组化法测定其ICAM-1的表达。结果:孕鼠肝脏组织ICAM-1表达,模型组与正常组相比差异有显著性意义(P<0·01);各治疗组与模型组相比差异有显著性意义(P<0·05或P<0·01);茵陈蒿高剂量组与正常组相比差异无显著性意义(P>0·05)。病理观察显示,经药物处理后孕鼠肝脏形态学改变得到不同程度改善。结论:茵陈蒿对于妊娠期肝内胆汁淤积症肝脏功能的改善有一定的效果,可能是通过调节肝脏组织ICAM-1的表达发挥其治疗作用。Objective: To explore the effects of Yinchenhao on liver pathohistology and intercellular adhension molecule-1 ( ICAM-1 ) of liver in pregnant rats with estrogen and progesterone induced intrahepatic cholestasis. Methods : The animal model of intrahepatic cholestasis of pregnancy rats was induced by estradioli benzoas and progesterone. All of rats were divided into norreal control group, model group, high close group, middle dose group, low dose group of Yinchenhao and ursodeoxycholic acid group at random. Pathohistological changes of the liver in all the rats were observed under light and electronic microscope. Moreover, ICAM-1 on liver was detected by immunohistochemis try staining. Results: In comparison of normal control group with the model group, there was significant difference in the expression of ICAM-1 ( P 〈 0. 01 ) . In comparison of all of the group under treatment with the model group, the expression of ICAM-1 was significantly changed ( P 〈 0. 05 or P 〈 0. 01 ) . High dose group of Yinchenhao had no difference with normal control group ( P 〉 0. 05 ) . The pathological changes showed different degrees of improvement were observed in histopathologic examinations of the liver in pregnanct rats under treatment. Conclusion: Yinchenhao is effective in the improvement of liver function of intrahepatic cholestasis of pregnancy and may be active by regulating the level of ICAM-1 on liver in ICP rats.
