动脉粥样硬化家兔主动脉中胶原和基质金属蛋白酶的变化及PACAP的影响

摘　　要：目的通过观察胶原和MMP-2在动脉粥样硬化（AS）家免主动脉中的变化，探讨AS过程中垂体腺苷酸环化酶激活肽（PACAP）的干预作用。方法雄性新西兰家兔60只，随机等分为正常对照组（C组）、AS模型组（AS组）和PACAP干预组（P组）。分别于实验的第4，8，12周末随机处死每组家兔各5—8只，取胸主动脉固定，常规石蜡切片，行HE染色、Masson染色及免疫组化染色，光镜下观察主动脉的组织学变化、胶原的变化和MMP-2的表达并利用图像分析系统对胶原进行半定量。结果与C组相比，AS组及P组主动脉斑块检出率及斑块内胶原容积分数处于上升趋势，MMP-2在斑块内阳性表达也呈增强趋势，但P组的斑块检出率、胶原容积分数及MMP-2的表达强度均低于同期AS组水平。结论PACAP可能通过抑制内膜中胶原沉积从而发挥抗AS的效应。Objective To observe the changes of collagen and MMP -2 in atherosclerosis and to study the effect of PACAP. Methods 60 male New Zealand rabbits were equally randomly divided into 3 groups： control group （C group） and atherosclerosis model group （AS group） and PACAP intervention group （P group）, At the 4th, 8th, 12th weekends of the experiment, 5 - 8 animals in each group were sacrificed, and arteries were harvested for paraffin sections. Sections were stained with hematoxylin - eosin and Masson separately, the distribution of collagen were examined and analyzed quantitatively with computer system and MMP - 2 was detected by immunohistochemisty. Results Comparing with the C group, the content of both collagen and percentage of plaque formation in AS group and P group increased in aortic intimae, the expression of MMP - 2 was increased in AS group and P group, but P group was lower than those in AS group. Conclusion PACAP may have good effect on AS by decrease of collage in aortic intimae.
