摘 要:目的观察脊髓损伤(SCI)后补阳还五汤是否有促进移植间充质干细胞(MSC)转分化为神经元作用。方法SD大鼠30只,以自制改良Allen装置造模成功后,随机分为模型组(不治疗)、补阳还五汤组、补阳还五汤加MSC组各10只。补阳还五汤组大鼠造模后当天即开始每天一次中药灌胃(3g/kg);补阳还五汤加MSCs组大鼠造模后随即移植MSC,细胞浓度1×106/mL,每鼠4.5!L,注入大鼠蛛网膜下隙,注射速度1!L/min,并开始每天一次中药灌胃,剂量3g/kg。4周后观察脊髓结构变化。结果与模型组相比,补阳还五汤组脊髓组织结构明显改善,前角运动神经元数量有所增加;补阳还五汤加MSC组结构最佳,并且前角运动神经元数量最多。结论由此推测补阳还五汤可能有促进移植MSC转分化为脊髓前角运动神经元作用。Objective To investigate the effect of Buyang-Huanwu decoction on transdifferentiation of MSC transplantation after spinal cord injury in rats. Methods The spinal cord injuried model was established by the modified Allen's method. 30 SD rats were randomly divided into three groups (10 rats/group): Model group(no treatment); Buyang-Huanwu decoction group which was administered daily; Buyang-Huanwu decoction and MSC transplantation group that received MSC transplantation and administed daily. By the end of the fourth week, the morphological features were evaluated by HE staining. Results compared with the model group, the tissue of spinal cord in the Buyang-Huanwu decoction group were recovered significantly; the Buyang- Huanwu decoction and MSC transplantation group was the best, and the anterior horn motor neurons in spinal cord were much more than those in the Buyang-Huanwu decoction group. Conclusion Buyang-Huanwu decoction might facilitate the transdifferentiation of MSC transplantation in the anterior horn motor neurons.
