摘 要:目的观察TnC基因在正常小鼠腭突发育和全反式视黄酸诱导的小鼠腭裂形成过程中的表达变化,探讨TnC基因的表达在腭裂发生中的作用。方法利用real-time PCR和原位杂交方法动态检测TnC基因在正常腭突组织和全反式视黄酸诱导的腭裂组织中的表达情况。结果real-time PCR结果表明,TnC在GD11-16正常腭突和腭裂组织中均有表达,在正常腭中第16天TnC表达丰度最高,与其他胚龄相比较差异有统计学意义。在腭裂组中的表达丰度明显低于正常腭,差异有统计学意义;而在腭裂组织中各胚龄TnC的表达丰度差异无统计学意义。原位杂交结果显示,TnC在GD13、GD14天正常腭组织的腭板前部上皮细胞和间充质细胞中均有表达,而在GD14天腭板后部上皮细胞和间充质中虽有表达,量较少;TnC在GD13天腭裂前部间充质细胞中的表达量明显下降,在GD14天的腭裂腭板前部和后部的间充质细胞和上皮细胞中均未检测到TnC的表达。结论TnC基因在维持小鼠腭突组织的生长发育中起重要作用,而在腭突组织中的抑制表达与腭裂的发生有关。Objective To observe the expression of TnC gene in development of normal palate shelf and the formation of cleft palate of mice.It is aimed to explore the relation between TnC gene and the formation of cleft palate.Methods The TnC expression in embyonic palatine process tissues in the experimental group induced by retinoic acid and control groups was detected by real-time PCR from GD11~GD16 and in situ hybridization at GD13 and GD14.Results TnC was expressed in all normal and cleft palates at GD11~GD16.To the normal palates,the highest expression was in GD16,which was significant differences to the other time samples.There was no difference of abundance among all the samples of the cleft palates.The TnC expression in normal palates was stronger than that in the cleft palates during GD11-GD16.TnC expression was detected in both the epithelium and mesenchyme of the anterior region of normal developing palatal shelves at GD13 and GD14,the TnC expression was detected in the the epithelium,but was down regulated in mesenchyme of GD13 palatal shelf.TnC was expression in the normal posterior palate at GD14,but was lower than its of anterior region.TnC was absent in both the epithelium and mesenchyme of the anterior region of cleft palatal at GD14.Conclusion TnC gene played an important role in development of palate shelf of mice.The down expression of TnC gene was related to the occurrence of cleft palate.
