摘 要:目的观察甲泼尼龙(MPL)对肺炎支原体(MP)诱导A549细胞产生细胞因子白细胞介素-8(IL-8)和肿瘤坏死因子-α(TNF-α)的影响,为临床在MP肺炎中合理应用糖皮质激素(GCs)提供理论依据。方法体外培养A549细胞和MP,用不同感染复数的MP诱导A549细胞,将0.015%的MPL加入到MP诱导的A549细胞中共同孵育,采用双抗体夹心ELISA法检测MPL作用于A549细胞后细胞分泌的IL-8和TNF-α的含量。结果MP能显著诱导A549细胞分泌IL-8和TNF-α,0.015%MPL能明显抑制A549细胞分泌IL-8和TNF-α(P<0.01),随着MP感染复数的增加,MPL的抑制作用降低。结论GCs可抑制MP诱导的A549细胞IL-8和TNF-α的分泌,提示在重症MP肺炎早期,适当应用合理剂量的GCs可减轻炎症反应的强度。Objective To observe the effect of methylprednisolone (MPL) on cytokine IL-8 and TNF-α secretion in A549 cells induced by Mycoplasma Pneumonia (MP). Methods A549 cells and MP were co-cultured in vitro. 0. 015% MPL was added into A549 cells which were pre-induced with MP at different multiplicity of infection (MOI). Enzyme-linked immunosorbent assay (ELISA) was used to detect the contents of IL-8 and TNF-α before and after MPL treatment, Results MP could significantly induce IL-8 and TNF-α secretion in A549 cells, The contents of IL-8 and TNF-α were greatly depressed after 0. 015% MPL treatment (P〈0.01), and the inhibitory effect of MPL was attenuated with the increase of MOI of MP. Conclusion Glucocorticoids can obviously inhibit the MP-induced A549 cells to secrete IL-8 and TNF-α, suggesting that the clinical application of gluococorticoids can attenuate the inflammatory response of severe MP in early stage.
