Effects of EphB/Ephrin-B1 Signal on the Currents of α3-nAChRs in Ciliary Ganglion Neurons

摘  要:目的探讨EphB/Ephrin-B1信号对鸡胚睫状神经节(ciliary ganglion,CG)神经元上α3-尼古丁乙酰胆碱受体(nicotinic acetylcholine receptors,α3-nAChRs)电流的影响。方法用膜片钳技术分别在培养的和急性分离的CG神经元上记录α3-nAChRs电流,细胞被随机分为:对照组;Ephrin-B1Fc治疗组——用Ephrin-B1与IgG重组后形成的Ephrin-B1Fc刺激细胞;IgG治疗组——用重组Ephrin-B1Fc所需相同浓度的IgG刺激细胞;Ephrin-B1治疗组——用重组Ephrin-B1Fc所需相同浓度的Ephrin-B1刺激细胞,观察电流的变化。结果对照组、IgG治疗组和Ephrin-B1治疗组之间α3-nAChRs电流无显著差异(P>0.05),而此3组与Ephrin-B1Fc治疗组之间有显著差异(均P<0.05),Ephrin-B1Fc可浓度依赖性抑制α3-nAChRs电流,该过程可快速发生并呈现可逆性。培养的和急性分离的CG神经元Ipeak/Cm的IC50分别为4.9μg/ml和8.4μg/ml。结论本研究表明Eph/Ephrin-B1信号可影响CG神经元α3-nAChRs的电流,提示Eph/Ephrin-B1可能参与交感神经系统功能的调控。Objective To investigate the effect of EphB/Ephrin-B1 signal on the currents of α3-nicotinic acetylcholine receptors (α3-nAChRs) on ciliary ganglion neurons. Methods Currents of α3-nAChRs on cultured and acute-associated ciliary ganglion neurons were recorded by using patch clamp technique. Cells were randomly divided into the following groups: control group, Ephrin-B1Fc-treated group (stimulating cells with different concentrations of recombinant Ephrin-B1Fc), IgG-treated group (stimulating cells with IgG), Ephrin-B1-treated group (stimulating cells with Ephrin-B1). The changes in currents were observed. Results There was no significant difference in the currents of α3-nAChRs between control, IgG-treated and Ephrin- B1-treated groups, but there was significant difference between these three group and Ephrin-B1Fc-treated group (all P〈 0. 05). Ephrin-B1Fc could concentration-dependently inhibit the currents of α3-nAChRs and the inhibitory effects happened quickly and were reversible. In the cultured and acute-associated ciliary ganglion neurons, IC50 of Ipeak/Cm was 4.9 and 8.4ug/ ml, respectively. Conclusion EphB/Ephrin-B1 signal can affect currents of α3-nAChRs on ciliary ganglion neurons, suggesting that EphB/Ephrin-B1 signal may play some important roles in regulation of sympathetic nervous system.
