摘  要:目的研究大鼠体神经-内脏神经及体神经-体神经吻合术后神经再生过程中,脑源性神经营养因子(BDNF)及其受体酪氨酸激酶B(TrkB)在脊髓前角运动神经元中的表达,比较两者有无差异,探讨相关因子在异类神经元再生过程中所起的作用。方法将45只雄性SD大鼠随机分为正常对照组、模型组、模型对照组。建立大鼠体神经-内脏神经吻合(模型组)及体神经-体神经吻合(模型对照组)模型,运用逆转录-聚合酶链式反应(RT-PCR)检测正常对照组、模型组和模型对照组术后7、14、30、60d脊髓腰4(L4)运动神经元中BDNF及TrkB mRNA的表达。结果在正常大鼠L4前角运动神经元中,BDNF及其受体TrkB均存在一定水平的表达。大鼠体神经-内脏神经吻合术后7、14、30、60d BD-NF和TrkB mRNA表达均升高,于术后7d达峰值,14d开始下降。大鼠体神经-体神经吻合术后BDNF和TrkB mRNA的表达水平逐渐增高,于术后14d达峰值。结论大鼠体神经-内脏神经反射弧建立后脊髓L4前角运动神经元BD-NF及TrkB的表达增高,BDNF及TrkB作为内源性保护性因子,其增高可能有利于受损神经元的存活和再生。大鼠体神经-内脏神经及体神经-体神经吻合术后BDNF及TrkB上调趋势存在一定的差异。Objective To study the differential expression of brain-derived neurotrophic factor (BDNF) and its receptor, tropomyosin related kinase B (TrkB), in rat spinal cord motor neurons after somatic-autonomic and somatic-somatic ventral root (VR) anastomosis and investigate their roles in foreign nerve regeneration. Methods Male SD rats were divided into three groups at random., normal control group, experimental group and experimental control group. In experimental group, the somatic-autonomic reflex pathway was established in rats by intradural microanastomosis of the left L4 ventral root to L6 ventral root. In experimental control group, the left L4 ventral root of the rats was transected and then the two sides were anastomosed immediately. Reverse transcription-polymerase chain reaction (RT-PCR) technique was applied to detect the mRNA expression changes of BDNF and TrkB in L4 motor neurons at the day 7, 14, 30 and 60 respectively. Results There existed mRNA expression of BDNF and TrkB in normal L4 motor neurons. The mRNA expression of BDNF and TrkB was increased at day 7, 14, 30, 60 after L4 L6- VRs anastomosis, reached the peak at day 7 and began to decrease at day 14. Nevertheless, in experimental control group, the mRNA expression of BDNF and TrkB was increased gradually and reached the peak at day 14. Conclusion BDNF and TrkB mRNA expression was increased in L4 ventral horn neurons of spinal cord after the establishment of the somatic-autonomic reflex arc. The increases in the BDNF and TrkB may benefit the survival and regeneration of the injured motor neurons. There is differential expression of BDNF and TrkB after somatic-autonomic and autonomic-autonomic nerves anastomosis.
