摘  要:通过分析亚抑制浓度的丁香酚对金黄色葡萄球菌全基因组表达谱的影响,研究了丁香酚的抗菌作用机制。用丁香酚(250μg.mL-1)处理金黄色葡萄球菌ATCC 25923细胞45 min后,应用Affymetrix金黄色葡萄球菌基因芯片检测全基因组表达谱的变化。共有338个基因受到诱导,440个基因受到抑制,其中包括100个转运子基因、45个与蛋白质合成有关的基因和32个毒力基因。丁香酚还抑制了大多数与细胞壁有关的基因和编码sortase酶的基因SA0982(srtB)以及Isd基因。结果表明,丁香酚是通过同时调节多途径相关基因表达,尤其是调节与转运、信号途径和毒力相关的基因表达来实现抗菌作用。To investigate the antimicrobial mechanism of eugenol against S. aureus, global transcriptional profiling of S. aureus in response to subinhibitory concentrations of eugenol were analyzed. S. aureus ATCC25923 was treated with 250 μg · mL^-1 eugenol for 45 min,Custom-made Affymetrix GeneChips^TM were used to determine the global transcriptional response of S. aureus ATCC25923 triggered by eugenol. Altogether 338 genes were identified to be upregulated and 440 genes were downregulated upon exposure to eugenol. Of these,100 transporter genes, 45 transcription regulator genes involved in protein synthesis and 32 virulence genes were differentially regulated by eugenol. Eugenol repressed the sortase-encoding gene SAO982(srtB) and lsd genes on cell wall. GeneChip analysis demonstrated that the antimicrobial mechanism of eugenol against S. aureus achieved through affecting different pathway genes simultaneously,especially genes involved in transportation, infection processes and information pathways.
