分光光度法测定盾叶薯蓣总皂苷的含量

摘　　要：目的用分光光度法测定盾叶薯蓣总皂苷的含量，为其质量控制提供依据。方法用分光光度法测定，以高氯酸为显色剂，显色温度70℃，时间15min，在407nm处有最大吸收。结果薯蓣皂苷在5．22—41．76μg／ml范围内呈良好的线性关系。精密度与回收率的RSD分别为1．9％和1，4％。方法学考察各项指标均符合要求。结论此法操作简便、准确、稳定性、重现性好。Objective： To apply a colorimetric analysis in the quantitative determination of total saponins and to offer the evidence for the quality control of Dioscorea zingiberensis C. H. Wright. Method： To use perchloric acid as chromogenic agent to determine total saponins, the heating temperature is 70℃ , the time is 15 min, and detect at the wavelength of the maximum absorption 407 nm. Results ： Dioscin showed a good linear relationship at a range of 5022 ～ 41.76μg/ml. RSD of the precision and recoveries were 1.9% and 1.4% respectively. All the indexes of the methodological investigation met the requirement. Conlusion： The method is simple and accurate. It has excellent stability and repeatability.
