摘  要:目的:观察刺蒺藜苷对慢性应激抑郁大鼠行为学及海马齿状回神经发生的影响,并探讨其机理。方法:采用慢性应激加孤养建立大鼠抑郁模型,通过敞箱实验等观察抑郁模型大鼠行为学改变,BrdU标记海马齿状回阳性细胞反映神经发生。结果:慢性应激抑郁大鼠体重增长缓慢(41.00±11.43g);蔗糖溶液消耗量减少(7.15±3.18g);敞箱实验中水平穿越格数(17.00±9.04格)、竖立次数(6.90±4.75次)、理毛时间有减少(4.56±1.71s),中央格停留时间(4.38±0.92s)、粪便粒数(9.34±3.01粒)均有增加。刺蒺藜苷和氟西汀可改善大鼠行为学变化,增加BrdU阳性细胞数(刺蒺藜苷1组10.23±3.81个;刺蒺藜苷2组10.92±2.16个;氟西汀组11.46±2.94个)。结论:刺蒺藜苷能明显改善抑郁动物的各项行为学指标,促进海马神经元发生,具有抗抑郁作用。Objective:To observe the effect of tribuloside on behavior and geurogenesis in dentate gyrus of chronic stress depression rats.Methods:All rats except the control group were singly housed and exposed an unpredicted sequence of mild stressor.The behavior of rats was detected by open-field test.And geurogenesis in dentate gyrus was observed by the Bromodeoxyuridine labelling method.Results:compared with control group,the weight of rats in model group showed significantly decreased(41.00±11.43g).Squares crossed(17.00±9.04),rears(6.90±4.75),and grooming(4.56±1.71s)in open-field test reduced,while time of staying in central square(4.38±0.92s)and defecation(9.34±3.01)were increased.And fluoxetine or tribuloside significantly improve behavioral disorder and BrdU immunoreactive cells(no.1 tribuloside group 10.23±3.81;no.2tribuloside group 10.92±2.16;fluoxetine group 11.46±2.94).Conclusion:tribuloside can improve behavior and BrdU immunoreactive cells of depression model rats and possess antidepressant effect.
