摘 要:目的:研究前摄宁颗粒(黄芩、黄柏、黄芪、党参、牡丹皮、丹参等)对大鼠前列腺炎的抗炎作用。方法:(1)采用角叉菜胶致大鼠急性非细菌性前列腺炎模型,雄性SD大鼠70只随机分成正常对照组,模型组,前摄宁颗粒大剂量组(6g/kg)、中剂量组(3g/kg)、小剂量组(1.5g/kg)和前列通阳性对照组(2.0g/kg),连续灌胃给药,第7天给药后1小时,1%角叉菜胶造模,测定外周血和前列腺液白细胞数、前列腺重量指数。(2)采用大肠杆菌致大鼠细菌性前列腺炎模型,动物分组及给药方式同角叉菜胶模型,于给药第4天造模,第7天给药后,测定外周血和前列腺液白细胞数、卵磷脂小体密度和前列腺湿重,摘取前列腺,进行解剖学观察和组织形态学观察。结果:(1)在急性前列腺炎模型中,前摄宁颗粒对前列腺湿重的增加有一定抑制作用,明显抑制前列腺液白细胞数的升高,减轻前列腺与周围组织黏连。(2)在细菌性前列腺炎模型中,前摄宁颗粒明显降低前列腺湿重,前列腺肿胀度抑制率达到48%~73%;明显升高前列腺液卵磷脂小体密度,降低前列腺液白细胞数;明显抑制前列腺组织间质增生。结论:前摄宁颗粒对大鼠慢性前列腺炎具有显著的抗炎作用,对急性非细菌性前列腺炎具有一定的抑制作用。AIM: To investigate the anti-inflammation effects of Qianshening Granules( Radix Scutellariae, Cortex Phellodendri Chinensis, Radix Astragali, Radix Codonopsis, Cortex Moutan, Radix et Rhizoma Salviae Mihiorrhizae, etc. ), a new preparation on SD rat prostatitis. METHODS : ( 1 ) Nonbacterial prostatitis caused by carrageenin was used to study the effect of Qianshening on acute prostatitis in rats. 70 SD rats were divided randomly into 6 groups: the normal control group, the positive control group(2.0 g/kg Qianlietong), the model group, the 6.0 g/kg Qianshening group, the 3.0 g/kg Qianshening group, and the 1.5 g/kg Qianshening group. All preparations were administrated by intra-gastric infusion, and 1 h after administration in the 7th day, 1% carrageenin was infused into one side of prostate. 4 h later, WBC in prostate liquid and in blood, and the prostate weight were detected. (2) Bacterial prostatitis caused by E. coli were used to study the effects of Qianshening on chronic prostatitis in rats. 67 SD rats were divided randomly into 6 groups: the normal control group, the positive control group(2.0 g/kg Qianlietong), the model group, the 6.0 g/kg Qianshening group, the 3.0 g/kg Qianshening group, and the 1.5 g/kg Qianshening group. All preparations were administrated by intra-gastric infusion. On the 4th day of administration, 100 μL of 1.0 × 10^7/mL E. coli was infused in one side of prostate. On the 7th day, WBC in prostate liquid and in blood, corpuscular densitys of lecithin, prostate weight, and histomorphology of prostate were detected. RESULTS: In the nonbacterial prostatitis model, Qianshening inhibited slightly the proliferation of prostate tissue, decreased the numbers of WBC in prostate liquid (P 〈0.05 in 3.0 g/kg Qianshening group, P 〈0.01 in 6.0 g/kg Qianshening group, comparing with the model group repectively) , alleviated the adherence of the prostate to the peripheral tissue.
