摘 要:目的优化葛根总黄酮提取工艺。方法采用水提壳聚糖澄清法与传统水提醇沉法平行对比,正交实验考察解吸剂(乙醇)浓度、解吸温度、洗脱速度3个因素,以总黄酮提取量、葛根素提取量、得膏率为考察指标综合评价。结果壳聚糖澄清工艺能有效保留葛根中葛根素及总黄酮等有效成分,节省乙醇用量,缩短生产周期。选取最佳纯化条件:乙醇浓度为70%,洗脱温度35℃,洗脱速度为2BV/h。采用该工艺葛根总黄酮纯度达60%以上。结论该法简单可取,适合于工业化大生产。Objective To optimize the extraction technology of the total flavones in Radix Puerariae. Methods Contrast the methods of clarifying water extractive solution of Radix Puerariae by chitosan and by alcohol. The orthogonal design method was adopted to study the effect of three factors including the concentration of the alcohol, the temperature and the speed of desorption. Extraction rate of total flavones and puerarin, the yielding rate of extractive were regarded as indexes. Results The clarifying process of chitosan could remain the effective contents such as the total flavones and puerarin. It not only saved the volume of alcohol, but also cut down the period of production. The optimum separation conditions were the elute solution of 70% alcohol, the speed of 2 drops per second and the temperature of 35℃. The yield of total flavones from Radix Puerariae by the purity reached above 60%. Conclusion The process is simple and preferable, which adapts to industrial production.
