摘  要:目的通过观察云芝多糖(CVP)对CCl4肝损伤抗氧化酶、自由基及NO含量的变化,探讨CVP抗肝损伤的作用机制。方法4月龄昆明种小鼠50只,分正常对照组、病理造模组、CVP保护组分高、中、低3组,常规饲料,自由饮水。CVP组每天按50,100,200 mg/kg体重的云芝多糖灌胃1次,饲养8 d,末次灌胃2 h,正常对照组腹腔注射调和油溶液,其余各组腹腔注射0.15%CCl4调和油溶液(10 ml/kg体重),12 h后禁食不禁水,18 h后处死小鼠,取出肝脏称重计算肝体指数;制备10%的肝匀浆,测定SOD,GSH-Px,NOS的活性和MDA,GSH,NO的含量。结果与模型组比较,CVP可降低肝体指数(P(0.05)和MDA的含量(P(0.001),提高抗氧化酶的活性和GSH含量(P(0.01),降低NOS活性和NO的含量(P(0.01)。结论CVP可减轻肝损伤所致炎性反应,提高抗氧化酶的活性和GSH的含量,加速自由基的清除,降低NO的量,对实验性肝损伤有保护作用。Objective To study the protective effect of Coriolus versciclor polysaccharides on CCl4 - induced liver injury in rats through observing the contents of antioxidase, free radicals and NO. Methods Fifty Kunming mice of 4 months old were divided into control group, model group, groups treated with small, middle and large dose of CVP. Gastric perfusion was done with 50 mg/kg, 100 mg/kg and 200 mg/kg respectively everyday. Rats were given regular diet, free water for 8 days. 2 hours after the last gastric perfusion, mixed -edible- oil solution was injected for control group through belly cavity, other groups were treated with 0.15% CCl4 mixed with mixed- edible- oil( 10 ml/kg). After 12 hours rats were absoluted diet and after 18 hours rats were sacrificed. The liver was taken out and weighed and the liver body index was calculated. 10% liver homogenate was prepared for test of the levels of SOD, GSH - Px, NOS, MDA, GSH and NO. Results Compared with model group, the liver body indexes of treatment groups were lowered( P 〈 0.05 ), MDA content was decreased ( P 〈 0.01 ), antioxidase activity and GSH content were increased ( P 〈 0.01 ), and NOS activity and NO content were decreased ( P 〈 0.01 ). Conclusion CVP can eliminate inflammatory reaction of liver injury, increase antioxidase avtivity and GSH content, scavenge free radicals, decrease NO content, and protect from liver damage.
