摘  要:目的研究模拟高+G_x 环境中猴舌黏膜上皮热休克蛋白质70(HSP-70)的表达。方法以9只雄性猕猴为对象,随机分为4组,对照组+1 G_x/300 s;实验1、2、3组,分别为+15 G_x/200 s、+18 G_x/165 s、+21 G_x/140 s。采用免疫组织化学方法,研究模拟高+G_x 环境作用下猴舌黏膜上皮细胞组织病理学改变及 HSP-70基因的表达情况,探讨其表达水平的改变与高+G_x环境的关系。结果组织病理学观察:实验组和对照组猴舌黏膜上皮细胞无明显变化;免疫组织化学观察:对照组舌黏膜上皮细胞核 HSP-70呈阴性表达或弱阳性表达。实验组舌黏膜上皮细胞核HSP-70着色明显,呈强阳性表达,不同高+G_x 组之间无明显差别。结论本研究表明模拟高+G_x 环境可引起猴舌黏膜上皮细胞 HSP-70基因表达增强。Objective To observe the expression of heat-shock protein 70 (HSP-70) in tongue mucous membrane epithelium in the stress reaction of rhesus macaque under +Gx loads. Methods Nine male rhesus macaques were randomly divided into 4 groups: control group was exposed to +1 Gx/300 s load, and experimental groups 1, 2 and 3 were exposed to + 15 Gx/200 s, +21 Gx/165 s and + 21 Gx/140 s respectively. Tongue tissue was fixed with 10% buffered formaldehyde and embedded with paraffin, and histopathological changes were observed under microscope. The expression of HSP-70 was detected by immunohistochemical PicTureTM method in the mucous membrane epithelium of tongue. Results Gross and histological analyses showed that no significant pathological changes were observed in the membrane of tongue mucous epithelium in different experimental groups of the animals. Immunohistochemical staining demonstrated that overexpression of HSP-70 was detected in the nuclei of the epithelium in the experimental groups, but no or slight positive staining was also observed in the nuclei of the epithelium in control animals. There was no significant difference in HSP-70 expression in different experimental groups. Conclusion Overload gravity can induce the enhanced expression of HSP-70 in the epithelium of tongue mucous membrane of the experimental monkeys.
