摘 要:目的观察高+Gx过载作用对猴颌下腺组织结构和c—jun基因表达的影响。方法9只雄性恒河猴,随机分为4组。对照组与实验1、2、3组受+Gx及其作用时间分别为1Gx/300s、+15Gx/200s、+18Gx/165s、+21Gx/140s。使用专用动物离心机,加载后的动物即刻处死、取材、制片、染色。用免疫组织化学方法观察猴颌下腺组织c—jun表达。结果组织病理学观察:对照组和实验组猴颌下腺组织均未见明显病理学改变。免疫组化观察:对照组颌下腺导管上皮细胞c—jun为弱阳性表达;实验组c—jun表达明显增强,阳性产物呈棕黄色颗粒状弥漫分布于颌下腺导管上皮细胞核,作用强度不同的+Gx组之间无明显差别。结论高+Gx环境中可引起猴颌下腺导管上皮细胞c—jun基因表达增强。Objective To observe the structural changes and c jun expression of submandibular gland in rhesusmacaqnl followimg high sustained +Gx loads. Methods Nine male rhesus macaques were randomly divided into four groups: control group underwent +1 Gx/300 s on animal centrifuge, and experimental groups 1, 2 and 3 underwent +15 Gx/200 s, +18 Gx/165 s and +21 G x/140 s respectively. The tissue of submandibular gland was fixed with 10% buffered formaldehyde and embed ded with paraffin, and histopathological changes were observed with microscope. The expression of c- jun in submandibular gland was detected by immunohistochemical staining (S-P method) Results gross inspection and histological observation showed that no significant pathological changes were ob served in the submandibular gland tissues in all 3 experimental groups of the animal. Immunohistochemical staining demonstrated that over-expression of c-jun was detected in the nuclei of the ductal cells in 3 experimental groups, and negative or slight positive staining was also observed in the ductal epithelium of the gland in control animals. There was no obvious difference of c-jun expression in different experimental groups. Conclusions High sustained +Gx can induce the over-expression of c jun in the submandibular gland of the experimental monkeys.
