摘 要:为给冬枣的遗传改良提供依据,以冬枣组培苗叶片为试材,研究了影响其叶片再生不定芽的主要因素,包括基本培养基、pH值、激素种类和浓度配比以及培养条件。结果表明:适宜冬枣叶片分化的基本培养基为MS,适宜的pH值为5.8~6.0,TDZ为诱导冬枣叶片不定芽再生的最佳激素;冬枣叶片不定芽再生的适宜培养基为MS+TDZ 1.0 mg/L+IBA 0.1 mg/L,平均再生率达到92.52%,平均出芽数4.64个。接种后暗培养14 d有利于叶片分化。In order to provide evidence for improving hereditary character of Ziziphus jujuba cv.'Dongzao',the factors affecting in vitro leaf regeneration were studied in Ziziphus jujuba cv.'Dongzao',including basic medium,pH,the variety and concentration of hormones and culture condition.The results indicated that suitable basic medium for differentiation was MS.The pH should be kept 5.8~6.0.The efficiency of TDZ in leaf regeneration was significantly higher than BA.The proper leaf regeneration medium was MS+TDZ 1.0 mg/L+IBA 0.1 mg/L.In this way,the regeneration rate reached 92.52% and the adventitious buds per leaf disc were 4.64.In addition,14 d pretreatment in dark was necessary to increasing the regeneration rate.
