摘 要:目的探讨不同时段大鼠哮喘模型肺组织α-平滑肌肌动蛋白(alpha-smooth muscle actin,α-SMA)在转录和蛋白水平的表达及川芎嗪对其的作用机制。方法以卵蛋白致敏制备大鼠哮喘模型,腹腔注射不同剂量川芎嗪、地塞米松及联合用药(川芎嗪+地塞米松)干预,分别在激发干预后的1周、2周取肺组织,采用逆转录聚合酶链反应(semi-quantitatively reverse transcription-polymerase chain reaction,RT-PCR)和免疫组织化学S-P方法研究肺组织α-SMA的表达。结果与正常组比较,在激发哮喘后1周肺组织α-SMA的表达显著升高(P<0.05),2周后表达更加显著(P<0.001);与哮喘模型组比较,各用药干预组α-SMA的表达均显著降低(P<0.001),且随着干预时间的延长降低幅度增大,川芎嗪大剂量干预组与地塞米松组相似,干预至两周时接近正常水平,二者降低的幅度均大于川芎嗪小剂量和联合干预组,联合用药干预组的效果优于川芎嗪小剂量组。结论哮喘肺组织α-SMA过量表达与哮喘的发生发展密切相关,并在早期即开始参与哮喘的发病。川芎嗪可在转录和蛋白水平抑制哮喘大鼠肺组织α-SMA的过度表达。[Objective] To observe the effect of Ligustrazine on expression of α-SMA in the lung tissue of rats with asthma. [Methods] Sprague-Dawley(SD) rats were randomly divided into 6 experimental groups: normal group (A), model group(B), Dexamethasone group(C), low-dose of Ligustrazine group(D), high-dose of Lignstrazine group (E), combinative therapy group(F). The asthmatic model was established by repeated inhalation of Gvalbulium. The expression of α-SMA in the lung tissue of rats was detected by semi-quantitatively reverse transctiption-polymerase chain reaction (RT-PCR) and means of immunohistochemical staining. [Results] There was little expression of α- SMA in the lung tissue of normal rats. The expressions of α-SMA increased 1 week after OVA challenge (P 〈0.001) and increased more significantly 2 weeks after OVA challenge compared with that in group A and the same group in 1 week (P 〈0.001). In group C, D, E and F, the contents of α-SMA were significantly lower than that in group B (P 〈0.001). After intervening 2 weeks the contents of α-SMA in group C and group E were approaching that in the normal group and lower than that in group D and group F. The effect in group F was better than that in group D. [Conclusion] Overexpression of α-SMA in asthmatic lung tissue is closely related to the onset and development of asthmatic. Ligustrazine could inhibit the development of asthma by the way of suppressing the expression of α-SMA in the levels of transcription and protein.
