应用胰蛋白酶裂解反相HPLC法分析rHuEPO肽图

摘　　要：胰蛋白酶降解rHuEPO后，用反相HPLC方法分析EPO降解后的产物（EPO肽图），HPLC色谱图显示了23个肽峰。发现同一单位研制的不同批制品肽图非常一致，并与美国Amgen公司的rHuEPO的肽图无论在峰数、峰型及保留时间上基本一致。该法具有重复性和重现性好，灵敏度高等特点，适用于rHuEPO肽图分析。The peptide map of rHuEPO after being degradated by trypsin was analysed by reverse phase-HPLC(RP-HPLC). A total of 23 peptide peaks were observed on the HPLC profile. Good identities were observed among the peptide maps of various batches of EPO prepared by the sarne manufacture,and the number and shapes of peptide peaks,as well as retention times were basically identical with the products from Amgen Corp. The method showed good repeatabi1ity, reproducibility and high sensitivity,and suitable for peptide mapping of rHuEPO.
