分泌性重组人粒细胞-巨噬细胞集落刺激因子药效研究

摘　　要：应用本所研制的重组人粒细胞-巨噬细胞集落刺激因子（rhGM—CSF），按0．012和0．024mg／kg剂量静脉注射家犬可明显抗由环磷酰桉所致的白细胞减少和血小板减少；按0．075、0．15和0．3mg／kg剂量皮下注射经60Co照射致白细胞减少症的小且有明显恢复作用：增加脾细胞集落、外周血白细胞、骨髓有核细胞及巨噬细胞数。其药效强度与进口升白能近似。The dogs with leukopenia and thrombocytopenia caused by cyclophosphamide were intravenously injected with the recombinant human granulocyte - macrophage colony stimulatlmg factor (rhGM-CSF ) prepared by the authors at the dosages of 0.012 and 0.024mg/kg. The result showed significant inhibition of rhGM -CSF to 1eukopenia and throntocytopenia. After being subcutaneously injected with the rhGM-CSFA,the numbers of splenocyte colonys, peripheral blood leukocytes, nucleated ce1ls in bone marrow and macrophages of the mice with leukopenia caused by irradiation with 60Co-γ were significantly increased. The curative effect of the rhGM-CSF was similar to that of imported products.
