应用柱层析法纯化Vero细胞乙型脑炎疫苗

摘　　要：浓缩Vero细胞乙脑疫苗经SepharoseCL－6B柱层析可去除大部分杂蛋白，再经SphacrylS－500HR层析可进一步去除比病毒大的杂质及残余小分子量杂蛋白。经两步凝胶过滤层析纯化后，疫苗总蛋白含量减少约99％，抗原比活性提高87～165倍，抗原回收率达30％～50％，残余牛血清和残余细胞DNA均符合WHO有关规程要求，提纯疫苗的效力达到或超过中国地鼠肾灭活乙脑疫苗和日本提纯鼠脑拔苗的参考苗。篢he purification of Japanese encephalitis (JE) vaccine prepared with Vero cells by 2 steps of column chromatographys was reported in this paper. After being purified by Sepharose CL-6B column chromatography, the most part of impure proteins were removed,and the residual impurities with the sizes of larger than viruses and the impure proteins with small molecu1ar weights were further removed by Sephacryl S-500 HR column chromatography. After being purified by the 2 steps of chromatographys,the total protein contents in the JE vaccine reduced by about 99%,the specific activity of the vaccine improved by 87~ 165 folds,the recovery rate of it reached 30% ~50%,and the residual calf serum and cell DNA contents in it fitted in with relevant WHO requirements .The potency of the purified vaccine was similar to or exceeded those of inactivated JE vaccine reference prepared with hamster kidney cells (China) and purified JE vaccine reference prepared with murine brain tissues (Japan).
